Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in the aquaculture production of this oyster species. Therefore a selective breeding program for resistance to bonamiosis has been undertaken since 1985 bu Ifremer, leading to the production of several selected oyster families. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters (injected with the parasite) to two tested families of oysters originating from the selective breeding program. Mortalities were checked daily, and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection to... |

The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. Secondly, by comparing the QTLs detected in this study with those previously published for O. edulis in similar experimental conditions, we identified several potential QTLs that were identical between the different families, and also... |

Alterations of chromosome number have been observed in the somatic tissue of Crassostrea gigas diploids and artificial polyploids bred in this species. Tetraploid animals with lower chromosome counts in some or all of their somatic tissue are considered undesirable as parents either for commercial triploids (produced via a cross with diploids) or for tetraploid breeding because (1) they may confer the tendency to lose chromosomes and revert to lower ploidy levels to their offspring or (2) their offspring could have a lower ploidy directly because of potential links between somatic and gametic chromosome loss. The present study evaluated the phenomenon in bi-parental tetraploid families bred from parents of differing ploidy quality, including a... |

We have identified quantitative trait loci (QTL) in the flat oyster (Ostrea edulis) for resistance to Bonamia ostreae, a parasite responsible for the dramatic reduction in the aquaculture of this species. An F-2 family from a cross between a wild oyster and an individual from a family selected for resistance to bonamiosis was cultured with wild oysters injected with the parasite, leading to 20% cumulative mortality. Selective genotyping of 92 out of a total of 550 F-2 progeny (i.e., 46 heavily infected oysters that died and 46 parasite-free oysters that survived) was performed using 20 microsatellites and 34 amplification fragment length polymorphism primer pairs. Both a two-stage testing strategy and QTL interval mapping methods were used. The two-stage... |

Background Because of its typical architecture, inheritance and small size, mitochondrial (mt) DNA is widely used for phylogenetic studies. Gene order is generally conserved in most taxa although some groups show considerable variation. This is particularly true in the phylum Mollusca, especially in the Bivalvia. During the last few years, there have been significant increases in the number of complete mitochondrial sequences available. For bivalves, 35 complete mitochondrial genomes are now available in GenBank, a number that has more than doubled in the last three years, representing 6 families and 23 genera. In the current study, we determined the complete mtDNA sequence of Ostrea edulis, the European flat oyster. We present an analysis of features of... |

Bonamiosis due to the parasite Bonamia ostreae has been associated with massive mortality outbreaks in European flat oyster stocks in Europe. As eradication and treatment are not possible, the control of the disease mainly relies on transfer restriction. Moreover, selection has been applied to produce resistant flat oyster families, which present better survival and lower prevalence than non-selected oysters. In order to better understand the mechanisms involved in resistance to bonamiosis, cellular and molecular responses of 2 oyster groups (selected oysters and wild-type oysters) were analyzed in the context of experimental injection and cohabitation infections. Cellular responses including non-specific esterases detection, ROS production and... |

Use of SNPs has been favored due to their abundance in plant and animal genomes, accompanied by the falling cost and rising throughput capacity for detection and genotyping. Here, we present in vitro (obtained from targeted sequencing) and in silico discovery of SNPs, and the design of medium-throughput genotyping arrays for two oyster species, the Pacific oyster, Crassostrea gigas, and European flat oyster, Ostrea edulis. Two sets of 384 SNP markers were designed for two Illumina GoldenGate arrays and genotyped on more than 1000 samples for each species. In each case, oyster samples were obtained from wild and selected populations and from three-generation families segregating for traits of interest in aquaculture. The rate of successfully genotyped... |

Summer mortalities of the Pacific cupped oyster Crassostrea gigas have been described in the literature for decades. One solution to overcome this problem would be to select animals more tolerant to summer mortalities, as this trait was shown to be highly heritable (Degrémont, 2003). The identification of markers associated to the resistance would allow to better understand this complex phenomenon and, eventually to establish markers assisted selection. Recently, two moderately dense genetic linkage maps have been established in the Pacific oyster based on microsatellites and AFLP markers (Hubert and Hedgecock, 2004; Li and Guo, 2004). In this context, the development and mapping of SNPs will increase the density of the linkage map and consequently the... |

Two oyster species are cultivated on the French coasts: the Pacific cupped oyster (Crassostrea gigas), which has been introduced from Japan in the 1970's after the disappearance of the Portuguese oyster Crassostrea angulala, and the European flat oyster (Ostrea edulis). During the second half of the 1960's, the latter suffered successively from two diseases caused by protozoan parasites: marteiliosis (also called "maladie des Abers") and bonamiosis. In France, its production dropped from 20,000 tian in the 1960's to 2,000 today. The impact of these two pathogens as well as the exploitation of the remaining stocks show the importance of the conservation of this species, the only natural European species, whose population is more and more fragmented. |

PCR-based DNA markers enable the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European fiat oyster (Ostrea edulis) populations aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Similar patterns of variation were found using allozymes, microsatellites and mitochondrial DNA, aIl which support an isolation by distance model. High variability was observed in the range of diversity between populations using a mitochondrial marker, a reflection of small effective population sizes in sorne locations. A 10fold quantitative difference was observed in Fst,... |
